ABS: A study was undertaken to study thermoregulated genes (either in an insertion library, or from recombinants from genetic crosses) in the psychrotrophic bacterium Pseudomonas fluorescens, by transcriptional fusions with bacterial luciferase genes as reporter, since there is little information regarding the effect of temperature on the physiology of cold-adapted bacteria. A total of 842 bioluminescent clones was assayed, and maximal bioluminescence at temperatures of 8, 17, or 28 C was measured. The bioluminescence during the various growth stages, e.g. stationary or exponential, was also recorded. These data indicated that about 39% of genes were thermoregulated, which is more than previous studies suggest. The authors suggest that this direct screening method might be a useful tool in the study of temperature-adaptation mechanisms.
